What do I use to connect two sections of Southwire "Titan" conduit to each other? 1.5 and 2 inch conduit sections underground.

I don't think you can connect two different sizes with out a box but they should have a coupling for same size connections

How to best make a hole for this, a drill bit? Step bit? What size hole?

This is designed to use with standard electrical boxes and options which almost all have precut holes with pop-outs. If you are trying to mount it in a non-standard location I would be careful about doing so if using this with electrical lines. However, if all safety is followed your best bet will be a drill. Measure the pop-outs on any electrical box you have access to to determine the right size. And be sure that whatever material you are mounting this Connector to is not thicker than the length of the threaded mounting area, including the nut and rubber washer, on the Connector.

Will this connector seal out moisture that may flow from the electrical box?

No. These are designed to keep out liquid at the inputs and outputs of electrical box’s, terminal boxes, junction boxes and switch/outlet boxes using same size conduit. You should ensure your electrical panel is water proofed.
