Will this work well if applied directly to the studs then 1 layer of drywall? Would this be more effective than 1/8" soundproofing tape?

The way to do that would be to decouple the drywall and the studs with resilient clips or channels. https://www.amazon.com/Green-Glue-Noiseproofing-Whisper-Clips/dp/B00HU5E1XA/ref=sr_1_10?camp=1789&creative=9325&keywords=%22resilient+sound+isolation+clip%22+1&linkCode=ur2&qid=1565794639&s=gateway&sr=8-10

can you use this on the back of an outdoor fence to muffle roadnoise?

This is not really meant for outdoor use.

Willl this work with an existing single wall construction house as an additional layer to reduce sound transmission?

I used it in conjuction with a second layer of drywall. It needs the second layer to spread the liquid over the surface of the first layer. Look it up on YouTube. There are a few instructional videos that explain the use rather well.

I'm adding a layer of drywall to a plaster wall. Do I need to paint primer on the plaster wall first?

No reason to in regards to the Green Glue with drywall. You do want to make sure the wall is smooth though, by sanding it down if necessary.
