what is the difference between system 4.0 and 5.0?

What year does this update go to?

2015 for most domestic. Euro and Asian vehicles 2014-ish. I don't work on those, so I can't say for certain...

We were given 3421-153 by otc. will this update from 3.0? anything else needed?

This is the same part number I purchased a few years ago (2016). Assuming it's the same product (flash card and CD), it will upgrade your system to 5.0 and license through 2015 for most domestic vehicles. Check the specs for the exact years (find a link in the other answered questions), I don't have mine in front of me right now. The software on the disc needs to be installed on a computer to do the update, and if I recall correctly, it says it needs to be Windows XP, but 7 worked. I upgraded that computer to 10, but didn't get a chance to try it, just haven't had the need...

Does this 5.0 upgrade cover Domestic, Asian and European and up to what model years?

The info on OTC's website is- •GENISYS '15 SUPER BUNDLE DOM/ASIAN/EURO •GENISYS 2015 DOM/ASIAN 2014 EURO LOYALTY KIT I haven't figured out what "Loyalty Kit" means, but this bundle upgraded my Evo to 2015 model years for the vehicles I've checked. One word of caution, I don't use it on many Eurpoean brands, and I haven't poked around to see where they stop being supported. If you want me to look into a particular make/model, I can fire it up and see what it says...
