Q: Can you plug headphones into the line out?

No you can’t but you can use headphones from your interface

Q: With this I can plug a tube amplifier "speaker out", directly into an audio interface?

Not “directly”, but you can run your amp into the load box, then another cable from the Captor into an audio interface and record the sound. That’s specifically why I bought it. Works well. I do need to add some EQ and a speaker IR, which can be done in a modeler or your DAW.

Q: Do you get added sustain if you turn it up while recording to your DAW?

If you turn it all the way up, you're going to get distortion. To get more sustain, you'd need to invest in better pickups.

Q: will this handle a peavey 6505 plus 120 watt amp head on full volume gain and headphone level sound?

Peavey 6505 outputs no more power than other 100w amplifiers, so it's fine to use. Peavey merely claims 30 watts per tube when just over half of that is actually being used for sound output.
