Q: What size is the opening is the downspout?

The back opening is 2.75" x 2-ish. Down spout fits into it fine, but it's oriented "landscape" not "portrait", so I had to get a converter. If you have a horizontal (landscape) downspout, you should be good to go!

Q: Opposite direction - how large is the mouth opening?

Its as big as the drain. Very well made and heavy. I would recommend purchasing it.

Q: What is this made out of?

I am not sure exactly what it is made of- some resin I suspect. BUT it is totally solid and withstands weather of any kind. Mine has been outside for over two years.
