Yes, it works with a UV filter on the lens. Just make sure screw the UV filter into your lens first before you mount the hood.

Does this fit the D3300 on a 35mm prime lens?

No, the bayonet is different. Look up your 35 mm prime lens and in the description it should give the part number for the lens that will fit your lens. Then do a search with that part number and should fit your lens.

Will the lense hood create a vignette on the 70-300mm lense when zoomed all the way out?

Unlikely. I only see a little vignette with my 18-55mm lens.
